South Africa began their campaign with a 1-1 draw against Namibia on Wednesday, with Rowen Human rescuing a point for the hosts in the second half.
Botswana, meanwhile, got off to a positive start after claiming a 1-0 win over Eswatini thanks to Lebogang Ditsele's solitary strike.
Coach Morena Ramoreboli is no stranger to Botswana football as he recently led Jwaneng Galaxy to the Bostwana Premier League title last season.
Bafana have beaten Botswana in their last three meetings, with two of those victories coming in the last two editions of the COSAFA Cup.
Botswana's last triumph over South Africa came in 2019, with the Zebras defeating their neighbours on penalties in the quarter-finals.
